Effect of Xiaochuanping powder on the inflammatory response and airway remodeling in asthmatic rats

چکیده
OBJECTIVE: To observe the effect of Xiaochuanping powder (XP), a traditional Chinese prescription for the treatment of cough and asthma, on serum concentrations of eosinophil cationic protein (ECP), tumor necrosis factor (TNF)-α, and interleukin (IL)-4, eosinophil counts, as well as expression of matrix metalloproteinase (MMP)-9, tissue inhibitor of metalloproteinase (TIMP)-1 in the lung tissues of asthmatic rats. METHODS: Sixty clean-grade Sprague−Dawley rats were divided randomly into six groups: normal control (NC), asthma model, Guilong Kechuanning (GK) group, as well as high-, intermediate-, and lowdose XP groups. Rats were sensitized with ovalbumin (OVA) to trigger asthma. Serum concentrations of ECP, TNF-α and IL-4, eosinophil counts, as well as expression of MMP-9 and TIMP-1 in lung tissues were evaluated using an immunofluorescence method. mRNA expression of MMP-9 and TIMP-1 was determined using real-time quantitative polymerase chain reaction. RESULTS: Compared with the asthma-model group, serum concentrations of ECP, TNF-α, and IL-4, and eosinophil counts decreased significantly in the highand intermediate-dose XP groups and GK group (all P < 0.01). Protein expression of MMP-9 and TIMP-1 decreased significantly in the highand intermediate-dose XP groups and GK group (all P < 0.01). Transcription of MMP-9 and TIMP-1 mRNA and the ratio of expression of MMP-9: TIMP-1 in lung tissue were significantly lower (P < 0.01). CONCLUSION: XP can reduce TNF-α secretion, suppress the infiltration / activation of eosinophils, reduce serum concentrations of ECP and IL-4, reduce the protein and mRNA expression of MMP-9 and TIMP-1 in lung tissue, and regulate the balance between expression of MMP-9 and TIMP-1. In these ways, XP alleviated the inflammation and remodeling of the airways. © 2018 JTCM. All rights reserved.

The preventive effect of Brassica napus L. oil on pathophysiological changes of respiratory system in experimental asthmatic rat
Objective: Asthma is an airway complex disease defined by reversible airway narrowing and obstruction, chronic airway inflammation, airway hyperresponsiveness, and tissue remodeling. The purpose of this study was to determine the effect of Brassica napus L. (B. napus) on airway pathologic changes in a rat model of asthma. Materials and Methods: Twenty-four rats were divided into 4 groups: contr...
متن کاملAirway Inflammatory Biomarker: Could It Tailor the Right Medications for the Right Asthmatic Patient?
Asthma is a heterogeneous disease, in which asthmatic patients present with different clinical phenotypes, variable endotypes, and different response to asthma medicines. Thus, we are faced with an asthma paradox; asthma is diagnosed subjectively by clinical history and treated with biologically active drugs. To solve this paradox, we need objective airway biomarkers to tailor the proper medica...

Asthma is a disease characterized by spontaneous contraction of the airways in response to a wide variety of endogenous and exogenous stimuli. Many asthma models are used to mimic the human asthma model in the literature. In order to better understand the role of the cannabinoid (CB) 2 receptor in the ovalbumin (OVA)-induced asthma model, a combination of both selective CB2 agonist (AM1241) and...
متن کاملInactivated Mycobacterium phlei inhalation ameliorates allergic asthma through modulating the balance of CD4+CD25+ regulatory T and Th17 cells in mice
Objective(s): Th2 response is related to the aetiology of asthma, but the underlying mechanism is unclear. To address this point, the effect of nebulized inhalation of inactivated Mycobacterium phlei on modulation of asthmatic airway inflammation was investigated. Materials and Methods: 24 male BALB/c mice were randomly divided into three groups: control group (Group A), asthma model group (G...
